Product Specific Information

The monoclonal antibody eBio4E3 reacts with an epitope shared by mouse CD21 (CR2) and CD35 (CR1). CD21 and CD35 are alternatively spliced transcripts from the Cr2 gene, resulting in cell-surface proteins of 145 and 190 kDa, respectively. They are expressed by mature B cells but not on thymocytes, peripheral T cells, erythrocytes, or platelets, and show little evidence of expression on macrophages.

CD21 acts as a receptor for complement component C3d and Epstein-Barr virus (EBV), and participates in B-cell activation through the B cell receptor complex with CD19 and CD81. Cr2-deficient mice display impaired inflammatory and humoral immune responses in vivo. The eBio4E3 and eBio8D9 clones bind different epitopes and do not cross-block each other.

Applications Reported: Suitable for flow cytometric analysis.
Applications Tested: Validated by flow cytometric analysis of mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Antibody titration recommended for optimal performance.

Target Information

CD21 (complement receptor 2, CR2) binds C3 complement fragments and plays roles in uptake and retention of immune complexes, memory B cell survival, and humoral immune response development. It also acts as a receptor for EBV and participates in prion targeting.

CD35 encodes a membrane glycoprotein found on peripheral blood cells, glomerular podocytes, and follicular dendritic cells. It binds C3b and C4b, regulating complement cascade activity. Variations in CD35 form the Knops blood group system, with alleles F and S differing by 8 exons. A soluble plasma form has been described.
