Product Specific Information
The HIB19 monoclonal antibody reacts with human CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all developmental stages except plasma cells. Follicular dendritic cells also express CD19. Together with CD21, CD81, Leu13, and MHC class II, CD19 forms a multimolecular complex associated with the BCR, mediating signaling that induces tyrosine phosphorylation, calcium flux, and B cell proliferation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated and tested by flow cytometric analysis of human mononuclear cells at 5 µL (0.5 µg) per test.
A test is defined as the amount of antibody that stains a cell sample in a final volume of 100 µL. Cell number may range from 10⁵ to 10⁸ cells/test.
Fluorochrome Information:
eFluor 450 is an alternative to Pacific Blue, excited at 405 nm and emits at 445 nm. Ensure instrument compatibility with Violet laser detection.
Filtration: 0.2 µm post-manufacturing filtered.
Target Information
CD19 is a member of the immunoglobulin superfamily with two Ig-like domains. It is expressed on 100% of peripheral B cells and may also appear on certain myeloid leukemia cells. CD19 acts as a critical regulator of B cell activation and proliferation, forming complexes with the B cell receptor to lower activation thresholds. Mutations lead to hypogammaglobulinemia, while overexpression results in B cell hyperactivity.
