Product Specific Information

The MFL3 monoclonal antibody reacts with mouse Fas (CD95) Ligand, a 40 kDa type II transmembrane glycoprotein and member of the TNF family expressed by activated T cells. Interaction with its receptor CD95 induces Fas-mediated apoptosis. The human FasL antigen is known to be cleaved by matrix metalloproteinases (MMPs), producing a 26 kDa soluble form; sensitivity for the mouse antigen to MMPs has not been reported.

Applications Tested:
Tested by flow cytometric analysis of mouse FasL-transfected cells. Recommended ≤0.25 µg/test in 100 µL final volume. Cell number may range from 10⁵–10⁸ cells/test. Optimal titration is recommended.

Target Information

CD178 (Fas ligand, FasL) is a type-II membrane protein with the N-terminus in the cytoplasm and the C-terminus extending extracellularly. Its receptor, FasR, is a type-I membrane protein of the TNF/NGF receptor family. CD178 induces apoptosis upon binding FasR, existing as membrane-bound (45 kDa) or soluble (26 kDa) forms, the latter generated via metalloproteinase cleavage.

CD178 is predominantly expressed in activated T-lymphocytes and NK cells, as well as immune-privileged tissues such as testis and eye. Expression is also observed in thymus, liver, ovary, lung, heart, and kidney. CD178-mediated apoptosis plays a key role in antiviral immune responses and immune regulation. Altered CD178 expression is associated with atherosclerosis, inflammatory diseases, AIDS, hepatitis, and cancer.
