Target Information
Tau는 신경세포 축삭에서 주로 발견되는 미세소관 결합 단백질로, 튜불린 중합을 촉진하고 미세소관을 안정화하는 역할을 합니다.
C-말단은 축삭의 미세소관에 결합하고 N-말단은 신경 세포막 구성요소에 결합하여, Tau가 두 구조 사이를 연결하는 단백질로 기능함을 시사합니다.
과인산화된 형태의 Tau는 알츠하이머병(AD) 뇌의 신경섬유 엉킴(neurofibrillary lesions)을 구성하는 주요 성분이며, 미세소관 결합 기능을 손상시켜 신경세포 퇴화를 유발합니다.
GSK-3β, PKA, cdk5, casein kinase II 등의 다양한 세린/트레오닌 키나아제가 Tau를 인산화합니다.
과인산화된 Tau는 알츠하이머병 외에도 Pick병, 전측두엽 치매, 피질기저변성증, 진행성 핵상 마비 등 다양한 신경퇴행성 질환에서 발견됩니다.
