Target Information
Tau는 주로 축삭(axon)에 존재하는 신경세포 미세소관 결합 단백질로, 미세소관의 중합과 안정화에 관여합니다.
C-말단은 축삭 미세소관과 결합하고, N-말단은 신경 세포막 구성요소와 결합하여 미세소관과 세포막 사이의 연결자(linker) 역할을 합니다.
과인산화된 Tau는 알츠하이머병(AD) 뇌에서 신경섬유 엉킴(neurofibrillary tangles)의 주요 구성 성분이며, 이는 미세소관 결합 기능을 손상시켜 신경세포 퇴화를 유발합니다.
Tau의 인산화는 GSK-3β, PKA, CDK5, casein kinase II 등 다양한 세린/트레오닌 키나아제에 의해 조절됩니다.
과인산화된 Tau는 알츠하이머병 외에도 Pick병, 전측두엽 치매, corticobasal degeneration, progressive supranuclear palsy 등 다양한 중추신경계 질환에서 발견됩니다.
