Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA).
CD45 is expressed by all hematopoietic cells except erythrocytes and platelets, and plays an important role in lymphocyte activation through its tyrosine phosphatase activity.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometry on normal human peripheral blood cells.
Use 5 µL (0.8 µg) per test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test.

Important Notes:

  • NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
  • Use Invitrogen LIVE/DEAD Fixable Dead Cell stains with NovaFluor dyes.
  • Use CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) for optimal staining.
    • Add 5 µL per sample to antibody cocktails/master mixes before combining with cells.
    • For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates utilize Phiton™ technology with engineered nucleic acid dye structures for optimized fluorescence, minimizing spillover and spread.
Excitation: 636 nm; Emission: 727 nm; Laser: 633–640 nm (Red).


Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells. It represents ~10% of lymphocyte surface proteins and is absent in non-hematopoietic tissues.
CD45 regulates Src-family kinases, T/B-cell receptor signaling, cytokine receptor signaling, and integrin-mediated pathways.
It exists in multiple isoforms due to alternative splicing, influencing lymphocyte subtype expression and physiological state.
CD45 is essential for immune cell activation, signaling, and survival regulation.
