Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45, also known as Leukocyte Common Antigen (LCA). CD45 is expressed by all hematopoietic cells excluding circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase enzymatic activity and plays an important role in activation of lymphocytes.

Each product contains:

  • 1 vial of NovaFluor conjugate
  • 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.8 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes.

CellBlox Plus Blocking Buffer Usage:
This buffer (Cat. No. C001T06F01) is required for optimal staining with NovaFluor conjugates.
Add 5 µL per sample to antibody cocktails/master mixes prior to combining with cells.
For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Technology:
NovaFluor conjugates are based on Phiton™ technology utilizing novel nucleic acid dye structures that enable engineered fluorescent signatures with minimized spillover and spread impacts.
Excitation: 636 nm; Emission: 756 nm; Laser: 633–640 nm (Red Laser)


Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of all surface proteins in lymphocytes.
It is absent on non-hematopoietic cells and tissues.
CD45 is critical for lymphocyte development and antigen signaling, regulating Src-family kinases and JAK kinases, and modulating integrin-mediated signal transduction, apoptosis, and immune functions.
Multiple isoforms exist due to alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
