Product Specific Information

The 2D1 monoclonal antibody reacts with all isoforms of human CD45 (Leukocyte Common Antigen, LCA). CD45 is expressed by all hematopoietic cells except circulating erythrocytes and platelets. The cytoplasmic portion of CD45 has tyrosine phosphatase activity and plays an essential role in lymphocyte activation.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Recommended use: 4 µL (0.6 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

Each NovaFluor conjugate or kit is shipped with CellBlox Blocking Buffer.
Use 5 µL per sample for optimal staining, including single-color compensation controls or antibody cocktails.

NovaFluor conjugates utilize Phiton™ technology with nucleic acid dye structures engineered for optimized fluorescent signatures minimizing spillover and spread.

Target Information

CD45 (LCA) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, comprising ~10% of lymphocyte surface proteins. Absent in non-hematopoietic cells, CD45 regulates Src-family kinases, T/B-cell receptor signaling, cytokine receptor pathways, and integrin-mediated survival signaling. Multiple isoforms arise via alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic regions.
