Product Specific Information

Specificity:
This antibody is specific for amino acids 38–51 (GDGMVPPGIEDKIT) of the platelet-aggregation-stimulating (PLAG) domain of mouse podoplanin (PDPN). It possesses high affinity and specificity for the MAP epitope tag (GDGMVPPGIEDK), derived from the PLAG domain of murine podoplanin.

Preparation:
This antibody was raised by immunising rats with a 14-residue synthetic peptide (mpp3851) corresponding to amino acids 38–51 of the PLAG domain of mouse podoplanin. Spleen cells were then harvested and fused to P3U1 cells to generate stable hybridomas.

Target Information

Podoplanin (PDPN, T1A, gp38, Aggrus) is a mucin-type 1 glycoprotein (40–43 kDa) expressed in various tumors and normal cells, especially lymphatic epithelial cells and follicular dendritic cells. It localizes in stromal cells of peripheral lymphoid tissue and thymic epithelial cells.
Podoplanin regulates lymphatic endothelium and maintains the morphology of podocytes. It acts as a ligand for CLEC-2, with expression correlated to tumor invasiveness and metastasis potential.
Podoplanin is involved in cell migration, metastasis formation, and tumor invasion. Its expression can be induced via Src activation of Cas and is also observed on Th17 cells. Dysfunction of podoplanin is linked to diseases such as lymphangioma and subglottic neoplasm. It has also been proposed as a marker for lung injury.
