Crystallins are separated into two classes: taxon-specific (enzyme) and ubiquitous. The ubiquitous class constitutes the major proteins of vertebrate eye lens, maintaining transparency and refractive index. Lens fiber cells lose nuclei during development, so crystallins are retained throughout life and remain stable.
Mammalian lens crystallins are divided into alpha, beta, and gamma families. Gamma-crystallins are monomeric, highly symmetrical proteins lacking connecting peptides and terminal extensions. They are differentially regulated after early development. Gamma-crystallin genes (γA–γD) and pseudogenes (γE–γG) are tandemly organized in a gene cluster. Mutations or aging can lead to cataract formation.
