Product Specific Information

The APB5 monoclonal antibody reacts with mouse CD140b (PDGFRB), the beta chain of the platelet-derived growth factor receptor. PDGFRB is a receptor tyrosine kinase forming dimers (α/α, β/β, or α/β) upon ligand binding, leading to phosphorylation of substrates. It is expressed in embryonic tissues and mesenchymal-derived cells of adult mouse tissues and plays a role in fibrous atherosclerotic lesion formation.

Applications Tested: Tested on NIH/3T3 cells at ≤1.0 µg/test (100 µL final volume; 10⁵–10⁸ cells/test). Optimal concentration should be titrated per assay.

Light Sensitivity: This tandem dye is sensitive to photo-induced oxidation; protect vial and samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-57) for up to 3 days at 4°C in the dark with minimal signal loss.

Target Information

PDGFRB is a receptor tyrosine kinase implicated in atherogenesis and cellular transformation by the SIS oncogene. It acts as a mitogen for mesenchymal-origin cells. The receptor forms homo- or heterodimers depending on the bound growth factor (PDGF α or β). The PDGFRB gene, located on chromosome 5, is adjacent to GM-CSF and M-CSFR genes, all potentially associated with 5-q syndrome. A translocation between chromosomes 5 and 12 fusing PDGFRB (CD140b) with ETV6 leads to chronic myeloproliferative disorder with eosinophilia.
