Description:
Monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, mesenchymal stromal cells, endometrial stromal cells, and certain tumors such as melanoma. Expression on circulating blood cells is limited to a subset of T cells. The protein is heavily glycosylated, with more than 50% of its mass derived from carbohydrates.

Cross-reactivity:
P1H12 antibody cross-reacts with mouse, rabbit, and canine, but not rat.

Applications Reported:
Validated for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested for flow cytometry of normal human peripheral blood cells. Recommended usage: 5 µL (0.125 µg) per test in a final volume of 100 µL. Cell number range: 10⁵–10⁸ cells/test.

Light Sensitivity:
Tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in:

Target Information

CD146 (MCAM):
An integral membrane glycoprotein of the immunoglobulin superfamily, expressed at cell-cell junctions in endothelial cells and various normal and malignant cells. Functions as a calcium-independent adhesion molecule and may act in signal transduction via Fyn kinase, influencing actin cytoskeleton assembly. Plays roles in endothelial cohesion, melanoma metastasis, and neural crest cell adhesion during embryonic development. CD146 also triggers tyrosine phosphorylation of FYN and PTK2, leading to transient intracellular calcium increase.
