Description:
ACK2 monoclonal antibody reacts with mouse CD117 (c-Kit receptor, Steel factor receptor, stem cell factor receptor). This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells and mast cells, playing a key role in proliferation, differentiation, and activation. ACK2 is reported to be a blocking antibody.
Applications Reported:
Suitable for flow cytometry, immunoprecipitation, Western blot, and immunohistochemistry of frozen tissue sections. Also used in functional assays (Functional Grade purified ACK2 recommended for functional assays).
Applications Tested:
Validated by flow cytometric analysis of mouse bone marrow cells. Recommended usage ≤0.125 µg per test (100 µL final volume). Cell number may range from 10⁵ to 10⁸ cells/test. Titration recommended for optimal performance.

Target Information
KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for mast cell growth factor (stem cell factor). It regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
Activation of c-Kit by SCF induces receptor dimerization and autophosphorylation, triggering downstream signaling pathways (Ras/ERK, PI3K, PLC-γ, JAK/STAT). KIT is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
