The YB5.B8 monoclonal antibody reacts with human CD117 (c-Kit), also known as Steel factor receptor or stem cell factor receptor. This 145 kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells and mast cells. The interaction of c-Kit and Steel factor promotes proliferation and differentiation of hematopoietic progenitor cells and mast cell development. CD117 is also expressed by melanocytes, playing a role in signaling and activation.

Target Information

KIT (c-KIT) is a proto-oncogene and a type III transmembrane receptor for mast cell growth factor (MGF, stem cell factor). KIT was first identified as the cellular homolog of the feline sarcoma viral oncogene v-kit. Together with its ligand, KIT regulates growth and activation of various hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.

Deregulation of KIT receptor tyrosine kinase by mutation D816V is a key focus in therapeutic strategies for neoplastic mast cell growth. c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract Cajal cells. Upon binding of SCF, c-Kit dimerizes, leading to receptor activation and autophosphorylation of tyrosine residues (e.g., Tyr703), enabling downstream signaling such as Ras/ERK, PI3K, PLC-γ, and JAK/STAT pathways. c-Kit receptor activation is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
