Product Specific Information

The FJK-16s antibody reacts with mouse, rat, dog, porcine, bovine, and cat Foxp3 (FORKHEAD BOX P3, SCURFIN, JM2).
Foxp3 is a 49–55 kDa transcription factor identified as the gene defective in scurfy (sf) mice. It is highly expressed in CD4+CD25+ regulatory T cells (Treg) and induces Treg phenotype in CD4+CD25– cells when ectopically expressed.

Immunoblotting mapped the epitope to amino acids 75–125 of mouse Foxp3. In humans, this region is alternatively spliced. The antibody is optimized for use with the Foxp3/Transcription Factor Buffer Staining Set (Product #00-5523-00). Other buffers are not recommended.

Applications Reported: Intracellular flow cytometric analysis.
Applications Tested: Intracellular flow cytometric analysis of mouse splenocytes using Foxp3/Transcription Factor Staining Buffer Set and protocol. Recommended usage ≤ 1 µg/test (100 µL final volume).
Cell number per test: 10⁵–10⁸ cells (empirical). Titration recommended for optimal performance.

Target Information

FOXP3 (Forkhead box protein 3) is a transcriptional regulator essential for immune homeostasis.
Expressed at high levels in CD25+CD4+ regulatory T cells, low levels in CD4+CD25– cells, and absent in CD4–CD8+ T cells.
Defects in FOXP3 cause scurfy phenotype in mice and IPEX syndrome in humans.
Transcript variants encoding different isoforms have been identified. FOXP3 expression in human breast and colon cancer cells is regulated by p53 in response to DNA damage.
