Target Information
Smoothened drosophila homolog (SMOH) is a serpentine protein belonging to the G-protein coupled receptor Fz/Smo family with 787 amino acids. It contains seven transmembrane domains, four glycosylation sites, and a putative extracellular amino terminus. Smoothened, activated by β-Arrestin 2 and GRK2, acts as a receptor for Sonic hedgehog (Shh) and positively regulates the hedgehog signaling pathway along with the inhibitor patched protein (PTCH). This pathway controls vertebrate development, cellular proliferation, and differentiation by activating transcription factor Gli. Mutations in Smoothened are associated with sporadic basal cell carcinoma of the skin and it is widely expressed in the heart and brain.
