Product Specific Information
Intact IgG appears on a non-reducing gel as a ~150 kDa band and upon reduction generates a ~25 kDa light chain band and a ~50 kDa heavy chain.
Recombinant rabbit monoclonal antibodies are produced using in vitro expression systems developed by cloning specific antibody DNA sequences from immunoreactive rabbits. Individual clones are screened to select the best candidates for production.
Advantages of recombinant rabbit monoclonal antibodies:
- Better specificity and sensitivity
- Lot-to-lot consistency
- Animal origin-free formulations
- Broader immunoreactivity due to larger rabbit immune repertoire
Target Information
IL-33 (Interleukin-33) is a 270 amino acid protein belonging to the IL-1 family, possessing an IL-1-like C-terminal domain. It functions both as a proinflammatory cytokine and as an intracellular nuclear factor with transcriptional regulatory properties.
IL-33 binds to and signals through IL1RL1/ST2, recruiting MYD88, IRAK1, IRAK4, and TRAF6, thereby activating NF-kappaB and MAP kinases. It promotes the production of TH2-associated cytokines (IL-4, IL-5, IL-13) and induces pathological changes in mucosal organs.
IL-33 is proteolytically converted to a mature form by CASP1 and is highly expressed in high endothelial venules of tonsils, Peyer’s patches, and mesenteric lymph nodes, but nearly undetectable in placenta.
Prolonged IL-33 treatment in mice results in eosinophilia, splenomegaly, and severe mucosal pathology. IL-33 also co-localizes with heterochromatin and exhibits transcriptional repressor activity, supporting its dual role as cytokine and nuclear factor.
