Target Information
Focal Adhesion Kinase (FAK)는 125 kDa 비수용체형 단백질 티로신 키나아제로, Src의 기질로 작용하며 인테그린 신호 전달의 핵심 요소입니다. 세포 확산, 분화, 이동, 사멸, 세포 주기 G1–S 전환 등에 중요한 역할을 합니다. FAK는 중심 촉매 도메인과 C-말단 꼬리를 가지며, 이는 세포가 인테그린 수용체를 통해 세포외기질에 부착하는 접착점(focal adhesion)에 위치하도록 합니다.
Fibronectin과의 결합 시 FAK 티로신 인산화가 증가하며, Grb2와 c-Src의 결합 및 ERK2 MAP kinase 활성화를 유도합니다. 다양한 isoform을 인코딩하는 전사 변이체가 존재합니다.
