Product Specific Information

This eyedio monoclonal antibody reacts with human indoleamine-2,3-dioxygenase (IDO, INDO, IDO1), an intracellular enzyme that catalyzes the degradation of tryptophan to kynurenines. IDO is expressed in various tissues and cells including macrophages, plasmacytoid dendritic cells, and tumor cells. Its expression can be induced by IFN-γ and other inflammatory stimuli, contributing to immune suppression via tryptophan depletion and T cell regulation.

  • Reported Applications: Intracellular staining followed by flow cytometric analysis.
  • Tested Applications: Pre-diluted and tested using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00).
  • Recommended Usage: 5 µL (0.25 µg)/test; suitable for 10⁵–10⁸ cells/test.
  • Excitation/Emission: 407 nm / 786 nm; Violet laser required.
  • Light Sensitivity: Protect from light to prevent photo-induced oxidation.
  • Fixation Compatibility: Compatible with IC Fixation Buffer or Fix/Lyse Solution; not compatible with methanol-based fixation.
  • Buffer Recommendations: Use Super Bright Complete Staining Buffer or Brilliant Stain Buffer to minimize non-specific polymer interactions.

Target Information

IDO1 is an intracellular heme-containing enzyme responsible for oxidative cleavage of the indole ring of tryptophan, serotonin, and melatonin, producing kynurenines. It is widely expressed in human tissues, macrophages, and dendritic cells. IFN-induced IDO1 plays a key role in antimicrobial defense and immune regulation during infection, pregnancy, autoimmunity, transplantation, and cancer.
