Product Specific Information

This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily containing an ITIM domain. It is expressed on subsets of γδ T cells, NK (CD56dim), CD8+ and CD4+ T cells, primarily by effector/memory phenotype cells that are short-lived but capable of immediate effector function. Cadherin-E, -N, and -R serve as ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation.

Clones 13F12F2 and 13A2 appear to recognize similar epitopes based on cross-blocking studies.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested on normal human peripheral blood cells. Use at 5 µL (1.0 µg) per test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test.

Brilliant Violet™ 786 (BV786) emits at 786 nm and is intended for use with cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific polymer interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark.
Note: BV786 is not compatible with methanol-based fixation.

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric type II transmembrane receptor with immunoreceptor tyrosine-based inhibitory motifs (ITIMs) interacting with SH2 domains of SHP-1 phosphatase. It is expressed on NK cells and certain T cells involved in tumor and virus-infected cell lysis, mediating humoral and cell-mediated immune responses.
