Product Specific Information

This 13F12F2 monoclonal antibody reacts with human killer cell lectin-like receptor subfamily G, member 1 (KLRG1), a type II transmembrane inhibitory receptor of the C-type lectin superfamily containing an ITIM domain. It is expressed on subsets of γδ T cells, NK (CD56dim), CD8+ and CD4+ T cells, primarily in effector/memory cells with immediate effector function. Cadherin-E, -N, and -R are ligands for KLRG1, and their interaction inhibits cytolytic activity and proliferation.

The clones 13F12F2 and 13A2 recognize similar epitopes based on cross-blocking studies.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (1.0 µg) per test (100 µL final volume).

Fluorophore Information:
Brilliant Violet™ 786 (BV786) emits at 786 nm and is intended for use on cytometers equipped with a violet (405 nm) laser. Ensure instrument compatibility.

Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
Sensitive to photo-induced oxidation; protect vial and samples from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark. BV786 is not compatible with methanol-based fixation.

Target Information

KLRG1 (Killer cell lectin-like receptor G1, MAFA, 2F1-Ag) is a homodimeric inhibitory receptor of the lectin-like type 2 transmembrane family containing ITIM motifs. These motifs interact with SH2 domains of protein phosphatases such as SHP-1. KLRG1 is expressed on NK cells and certain T cells involved in tumor and virus-infected cell lysis and immune response mediation.
