Description:
The MP6-XT22 antibody reacts with mouse tumor necrosis factor-alpha (TNF-α), a 17 kDa cytokine produced by monocytes, macrophages, neutrophils, NK cells, and CD4⁺ T cells. TNF-α exhibits cytolytic activity against various tumor cells and plays a key role in immune regulation. It forms dimers and trimers and also exists as a 26 kDa membrane-bound form.

Applications Reported:
Reported for use in ELISA, neutralization, and immunohistochemistry.

Applications Tested:
The Functional Grade Purified MP6-XT22 antibody has been tested by LAL assay for low endotoxin levels and for neutralization of TNF-α bioactivity.
At 1.25 µg/mL, it neutralizes 50% of the biological effects of 1.0 ng/mL mouse TNF-α in an assay of TNF-α-induced cytotoxicity of L929 cells.

Recommendations:
Careful titration is recommended for optimal performance in the assay of interest.

Target Information

TNF-α is a multifunctional proinflammatory cytokine belonging to the TNF superfamily. It is mainly secreted by macrophages and binds to receptors TNFRSF1A/TNFR1 and TNFRSF1B/TNFBR. TNF-α regulates immune cell activity, cell proliferation, differentiation, apoptosis, lipid metabolism, and coagulation.
It forms multimers (dimers, trimers, or pentamers) and appears as a 17 kDa band under non-reducing SDS-PAGE conditions.
Knockout studies in mice indicate TNF-α’s neuroprotective functions and its role in tumor necrosis. It is involved in immune responses to bacterial, viral, parasitic, and fungal infections, and implicated in autoimmune diseases, insulin resistance, and cancer.
