Product Specific Information

The MP1-22E9 antibody reacts with mouse granulocyte/macrophage colony-stimulating factor (GM-CSF) and functions as a neutralizing antibody. Mouse GM-CSF (~14 kDa) is produced mainly by activated T cells and macrophages, and also by endothelium and fibroblasts in response to TNF-α, IL-2, IL-1, and IFN-γ. GM-CSF promotes growth of macrophages, granulocytes, and dendritic cells, existing both as membrane-bound and extracellular matrix-associated forms. Non-glycosylated GM-CSF is biologically active.

Applications Reported:
ELISA, ELISPOT, neutralization, and intracellular staining for flow cytometric analysis.

Applications Tested:
The Functional Grade Purified MP1-22E9 antibody has been tested by LAL assay (low endotoxin), ELISA, and bioassay for GM-CSF neutralization.
At 1.25 µg/mL, the antibody inhibits by 50% the effects of 1 ng/mL mouse GM-CSF in MC/9 cell proliferation assay.

ELISA Capture Use:
Used as capture antibody in sandwich ELISA with biotin MP1-31G6 (13-7332) for detection and recombinant mouse GM-CSF (14-8331) as standard.
Recommended capture concentration: 1–4 µg/mL.
Standard curve range: 1000 pg/mL – 8 pg/mL.

Target Information

The GM-CSF gene encodes a cytokine regulating production and function of macrophages and granulocytes. The active protein forms a homodimer in extracellular space. Located on chromosome 5q31, this gene cluster is associated with 5q- syndrome and acute myelogenous leukemia, alongside IL-4, IL-5, and IL-13.
GM-CSF promotes tissue inflammation; elevated levels have been observed in SARS-CoV-2 patients with acute respiratory distress syndrome.
Mice lacking GM-CSF or its receptor develop pulmonary alveolar proteinosis.
