Product Specific Information

The MD-1 antibody reacts with human, rhesus, cynomolgus, and baboon interferon-gamma (IFN-γ), a signature Th1 cytokine produced mainly by activated T cells, NK cells, and B cells. IFN-γ is a dimeric protein with antiviral and antiparasitic activities and inhibits proliferation of normal and transformed cells.

Applications Reported:
Intracellular staining, flow cytometry, cytokine neutralization, immunohistochemistry, ELISA capture, and ELISPOT capture.
Fluorochrome-conjugated 4S.B3 antibody is recommended for intracellular staining and flow cytometry.

Applications Tested:

  • ELISA Capture: Tested as a capture antibody for human IFN-γ, used with biotinylated 4S.B3 (13-7319) for detection and recombinant IFN-γ (39-8319) as standard. Recommended range: 1–4 µg/mL.
    Include a standard curve (1000 pg/mL – 8 pg/mL) for each plate.
  • ELISPOT Capture: Tested with biotinylated 4S.B3 antibody; suitable range 1–4 µg/mL.
  • Neutralization: At 30 µg/mL, neutralizes 50% of the effect of 1 ng/mL human IFN-γ. Clone NIB42 is more potent.

Storage and Handling:
Use in a sterile environment.

Filtration: 0.2 µm post-manufacturing filtered.
Purity: >90% (SDS-PAGE).
Endotoxin Level: <0.001 ng/µg (LAL assay).
Aggregation: <10% (HPLC).


Target Information

IFN-γ (Interferon gamma, Type II interferon)
A macrophage activation factor produced primarily by T-lymphocytes and NK cells in response to antigens and cytokines.

  • Structure: Dimeric glycoprotein (~45 kDa), composed of two 146 amino acid subunits.
  • Functions: Antiviral and antiproliferative activity, induction of MHC I/II, macrophage activation, and enhanced immunoglobulin secretion.
  • Species specificity: Human IFN-γ does not cross-react with mouse IFN-γ.
  • Regulation: Upregulated by IL-2, FGF basic, EGF; downregulated by vitamin D3 or DMN.
  • Associated with cytokine regulation and JAK-STAT pathway activation.
  • Mutations linked to aplastic anemia.
