This NJTEN3 monoclonal antibody reacts with the pro-form of mouse IL-1 beta, a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. The cytokine is synthesized as a 31 kDa inactive precursor that accumulates in the cytosol and is cleaved into the active 17 kDa form upon inflammasome activation. IL-1β lacks a classical secretion signal and is secreted alongside caspase-1 through an alternative mechanism.

Elevated IL-1β levels are linked to chronic inflammatory conditions, making IL-1β and its receptor antagonist (IL-1RA) important therapeutic targets. The NJTEN3 antibody specifically recognizes the pro-form of mouse IL-1β and does not bind the active (cleaved) form.

Applications Reported: Intracellular staining and flow cytometric analysis.
Applications Tested: Tested using mouse thioglycolate-elicited peritoneal macrophages with the Intracellular Fixation & Permeabilization Buffer Set (Product # 88-8824-00).
Use ≤0.06 µg per test in 100 µL final volume. Cell number range: 10⁵–10⁸ cells/test. Antibody titration recommended.

Target Information

Interleukin-1 beta (IL-1β) is a proinflammatory cytokine expressed by monocytes, macrophages, and dendritic cells. Synthesized as a 31 kDa inactive pro-form, it becomes active (17 kDa) through inflammasome-mediated caspase-1 cleavage. IL-1β signals via IL-1RI and IL-1RII, shared with IL-1α, and plays a key role in innate immune defense by inducing proinflammatory cytokines and acute-phase responses. Its activity is regulated by IL-1RA, which competitively inhibits receptor binding. Elevated IL-1β levels are associated with chronic inflammatory diseases.
