Target Information
The p90 ribosomal S6 kinases (RSKs) comprise a family of serine/threonine kinases at the terminus of the ERK pathway. In humans, the RSK family consists of four isoforms (RSK1–4). These kinases contain two distinct catalytic domains activated sequentially by phosphorylation.
RSK regulates gene expression via phosphorylation of transcriptional regulators such as c-Fos, estrogen receptor, NFkappaB/IkappaB alpha, and CREB. ERK activates the C-terminal kinase of RSK, which in turn activates the N-terminal kinase. RSKs are located in both cytoplasm and nucleus, and upon activation, cytosolic RSK translocates to the nucleus to phosphorylate transcription factors.
