Product Specific Information
The eBio4B10 monoclonal antibody reacts with mouse and human T-bet, a Th1-specific T-box transcription factor critical for Th1 CD4+ T cell lineage development.
T-Bet expression is induced by IFN-γ and regulates its own expression along with IL-12Rβ and IL-2, also contributing to class-switch recombination in B cells.
Applications Reported: Suitable for intracellular staining and flow cytometric analysis.
Applications Tested: Validated for intracellular staining and flow cytometry of human peripheral blood cells at ≤ 0.5 µg/test (100 µL final volume).
Optimal antibody titration is recommended per assay.
Use with Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and refer to BestProtocols® for intracellular staining (Protocol B).

Target Information
T-bet (TBX21) is a T-cell specific transcription factor containing a single T-box DNA-binding domain. It regulates expression of interferon-gamma and directs Th1 lineage development.
T-bet cooperates with Stat4 to program chromatin architecture for Th1 gene expression. Genetic variations are associated with aspirin-induced asthma and nasal polyps.
