Product Specific Information

The eBioMLC-51 monoclonal antibody recognizes mouse Nanog, a homeobox transcription factor essential for maintaining pluripotency in embryonic stem cells. Nanog expression is specific to early embryos, the inner cell mass of the blastocyst, ES cells, and EG cells, and is downregulated upon differentiation.

Nanog acts as a transcriptional activator with two activation domains (CD2, WR) in the C-terminus and one in the N-terminus. Immunoblotting with eBioMLC-51 shows a ~45 kDa band in F9 cell lysates but not in NIH3T3 or spleen lysates. For intracellular staining, use the eBioscience Foxp3 Staining Buffer Set (Product #00-5523).

Applications Reported: Intracellular staining with flow cytometry, western blotting, microscopy, immunohistochemistry, and immunocytochemistry.
Applications Tested:

  • ICC: ≤10 µg/mL
  • Flow cytometry: ≤0.06 µg/test using Foxp3/Transcription Factor Staining Buffer Set.
    A test is defined as the antibody amount used in 100 µL final volume, typically 10⁵–10⁸ cells/test. Optimize antibody concentration for best performance.

Target Information

NANOG (Nanog homeobox) is a transcription factor regulating pluripotency and differentiation in embryonic stem cells. NANOG mRNA is expressed in pluripotent cell lines and absent in differentiated cells. Human NANOG shares 52% overall identity with mouse NANOG and 85% in the homeodomain.
Human NANOG maps to 12p13.31; mouse NANOG to 6F2. NANOG expression occurs in the inner cell mass of the blastocyst. It promotes self-renewal and proliferation of ES cells and is implicated in iPS cell generation with Sox2, Klf4, and c-Myc expression. NANOG dysfunction is linked to teratocarcinoma and germ cell cancers.
