Product Specific Information
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain) to form the IL-7 receptor (IL-7R), a type I glycoprotein (75–80 kDa) expressed on immature B cells, thymocytes, and most mature T cells. IL-7 binding activates signal transduction through Jak/STAT and other kinase pathways, essential for lymphocyte development and T-cell homeostasis.
CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). CD4⁺CD25⁺CD127⁻ cells are enriched for Foxp3⁺ Tregs with suppressive activity, providing a more accurate Treg identification marker.
Binding of eBioRDR5 to PBMCs is blocked by recombinant human IL-7 (Product #14-1079-80).
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
- Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.
- Use CellBlox Plus Blocking Buffer (Product #C001T06F01) for optimal staining. Add 5 µL per sample before combining with cells.
- For single-color controls: 5 µL per 100 µL cell suspension (10³–10⁸ cells).
NovaFluor conjugates utilize Phiton™ technology, engineered for precise fluorescence spectra with minimal spillover and spread.

Target Information
CD127 (Interleukin-7 Receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. The IL7R complex (alpha/gamma heterodimer) mediates IL-7–induced signaling. The gamma(c) chain activates signal transduction, while the alpha chain determines specific pathways. CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature T cells. Dysregulation is associated with severe combined immunodeficiency (SCID).
