Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human and canine Ki-67 protein, expressed in dividing cells (G1, S, G2, and M phases) but absent in resting cells (G0). Ki-67 localizes to the nucleus during interphase and redistributes to chromosomes during mitosis. It associates with heterochromatin-bound proteins such as CBX3 and serves as a marker for proliferating cells in tumor studies.

Applications Reported:
Intracellular staining followed by flow cytometric analysis.

Applications Tested:
Pre-diluted and tested using the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00). Recommended use: 5 µL (0.25 µg) per test in a 100 µL final volume. Suitable for 10⁵–10⁸ cells/test.

Fluorochrome Information:
Brilliant Violet™ 605 emits at 605 nm and is compatible with cytometers equipped with a violet (405 nm) laser. Use Brilliant Stain Buffer™ (Product #00-4409-75) or Super Bright Complete Staining Buffer (Product #SB-4401-42) when combining multiple polymer dyes to minimize non-specific interactions.

Handling and Fixation:

• Protect from light (photo-induced oxidation sensitivity).
• Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
• Not compatible with methanol-based fixation.
• Excitation: 407 nm / Emission: 605 nm / Laser: Violet.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases of the cell cycle. It associates with euchromatin and heterochromatin regions and serves as a key proliferation marker since it is absent in G0 cells. Ki-67 expression correlates with tumor grade and is used to determine the growth fraction in neoplasms. Commonly applied as a neuronal marker of cell cycling and proliferation.
