Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8b chain associates with the CD8a chain to form the CD8ab heterodimer expressed on most thymocytes and peripheral cytotoxic αβ TCR T cells. CD8 receptor complex binds to MHC class I and plays a role in T cell development and activation.

SIDI8BEE detects CD8β in both CD8αβ heterodimers and CD8ββ homodimers.

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted; tested on normal human peripheral blood cells at 5 µL (0.25 µg)/test.
Cell sample volume: 100 µL; cell number: 10⁵–10⁸ cells/test (empirical determination required).

Fluorophore Information:
Brilliant Violet™ 605 (BV605) emits at 605 nm; use with violet (405 nm) laser-equipped cytometers.
Use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) when combining multiple polymer dyes to minimize non-specific interactions.

Light Sensitivity: Protect from light to prevent photo-induced oxidation.
Fixation Compatibility: Compatible with IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54); store up to 3 days at 4°C in dark.
Not compatible with methanol-based fixation.

Target Information

The CD8B antigen is a cell surface glycoprotein found on most cytotoxic T lymphocytes, mediating efficient immune cell interactions. CD8 acts as a coreceptor with the T-cell receptor to recognize antigens presented by class I MHC molecules. The functional coreceptor can be a homodimer (αα) or heterodimer (αβ). Both chains share homology with immunoglobulin variable light chains. The CD8B gene encodes the β chain isoforms, with multiple alternatively spliced variants described. A pseudogene on chromosome 2 has also been identified.
