Product Specific Information

The M5/114.15.2 monoclonal antibody reacts with mouse MHC class II molecules encoded by I-A and I-E subregions (I-A b, I-A d, I-A q, I-E d, I-E k, not I-A f, I-A k, or I-A s). It detects polymorphic determinants on B cells, monocytes, macrophages, dendritic cells, and activated T lymphocytes from mice carrying the H-2 b, H-2 d, H-2 q, H-2 p, H-2 r, and H-2 u haplotypes, but not H-2 s or H-2 f.

Applications Reported: Flow cytometric analysis.

Applications Tested: Tested by flow cytometric analysis of mouse splenocytes. Recommended ≤0.25 µg/test (100 µL final volume). Cell number may vary (10⁵–10⁸ cells/test). Optimize titration for best performance.

Brilliant Violet™ 605 (BV605) emits at 605 nm and is compatible with violet (405 nm) laser-equipped cytometers. Ensure instrument compatibility.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Light sensitivity: Protect vial and samples from light.

Fixation: Samples can be stored in IC Fixation Buffer (00-8222-49) or 1-step Fix/Lyse Solution (00-5333-54) for up to 3 days at 4°C in the dark. BV605 is not compatible with methanol-based fixation.

Target Information

MHC class II molecules are transmembrane glycoproteins expressed on antigen-presenting cells (macrophages, dendritic cells, B cells). They bind exogenous antigens and present them to T cells. The antigen-binding groove is open-ended (15–24 amino acids long).
HLA and MHC antibodies are essential in Immunopeptidomics for neoantigen identification via LC-MS/MS.
