• Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Desmin (approximately 53 kDa) exhibits high tissue specificity, being predominantly expressed in all muscle cell types (cardiac, skeletal, and smooth muscle).
Its expression is stage- and tissue-specific, induced during terminal skeletal muscle differentiation.
In skeletal and cardiac muscle cells, desmin is localized at the Z-disk and intercalated disk regions.
In smooth muscle, expression is more heterogeneous.
Coexpression of vimentin and desmin has been observed in muscle-derived tumors such as rhabdomyosarcomas and leiomyosarcomas.
During myocardial dysfunction, dramatic changes in desmin distribution have been observed.
