Product Specific Information
- Immunohistochemistry (PFA fixed):
Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
Desmin (approximately 53 kDa) exhibits a high degree of tissue specificity, being predominantly expressed in all types of muscle cells (cardiac, skeletal, and smooth muscle).
Its expression is stage- and tissue-specific, induced during terminal skeletal muscle differentiation.
In skeletal and cardiac muscle cells, desmin localizes to the Z-disk region and intercalated disks, while in smooth muscle its expression is more heterogeneous.
Coexpression of vimentin and desmin has been observed in tumors derived from muscle tissue such as rhabdomyosarcomas and leiomyosarcomas.
During myocardial dysfunction, dramatic changes in desmin distribution have been observed.
