Specificity: OTH-74D4 antibody recognizes CD45RA, an isoform of CD45 containing only the protein A region. The CD45RA antigen is expressed mostly on naive CD4-positive, CD8-positive, and γδ-positive T cells, as well as on most B and NK lymphocytes. CD45RA antigen density generally decreases upon activation.

CD45RA is an isoform of the CD45 complex with restricted expression among different lymphoid cell subtypes.
CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed in all nucleated hematopoietic cells, comprising about 10% of lymphocyte surface proteins.
It plays a critical role in lymphocyte development and antigen signaling by regulating Src-family kinases.
CD45 isoforms arise from alternative splicing, differing in extracellular domains but sharing identical transmembrane and cytoplasmic domains.
These isoforms vary in their ability to localize within glycosphingolipid-enriched membrane domains, depending on cell type and physiological state.
CD45 also promotes cell survival via integrin-mediated signaling and participates in DNA fragmentation during apoptosis.
It is expressed in human hematopoietic lineage cells except mature red cells, and not detected in other differentiated tissues.
CD45 dysfunction is associated with diseases such as severe combined immunodeficiency and Hepatitis C.
