Target Information
CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein belonging to the scavenger receptor cysteine-rich superfamily. It acts as a receptor for the hemoglobin-haptoglobin complex and helps protect tissues from oxidative damage caused by free hemoglobin. CD163 is involved in iron recycling through endocytosis and heme breakdown.
CD163 is expressed on human monocytes and macrophages, particularly during the late phase of inflammation. It is found on all circulating monocytes and most tissue macrophages, except those in mantle zones and germinal centers of lymphoid follicles. Expression is upregulated by IL-10, IL-6, and dexamethasone, while LPS and PMA induce shedding of CD163 from the cell surface.
CD163 binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, showing higher affinity for multimeric haptoglobin (HP1F) than for dimeric haptoglobin (HP1S). It triggers intracellular signaling cascades involving tyrosine kinase-dependent calcium mobilization, inositol triphosphate production, and secretion of IL6 and CSF1.
