Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa selectin family member expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. CD62L mediates lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Usage Notes:

• Use 5 µL (0.2 µg) per test (100 µL final volume).
• Cell number range: 10⁵–10⁸ cells/test.
• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.

CellBlox Plus Blocking Buffer (Cat. No. C001T06F01):

• Required for optimal staining with NovaFluor conjugates.
• Add 5 µL per sample regardless of number of NovaFluors.
• For single-color controls: 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor Conjugates:

• Based on Phiton™ technology using nucleic acid dye structures.
• Engineered to minimize spillover and spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor.
It consists of:

• Amino-terminal C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on lymphocytes, monocytes, and polymorphonuclear cells, excluding a substantial population of memory T cells.
