Product Specific Information

The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds glycosylated, fucosylated, and sulfated sialylated glycoproteins including CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Use 5 µL (0.4 µg) per test (final volume 100 µL, 10⁵–10⁸ cells/test).

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., PI, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability analysis.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining with NovaFluor conjugates.
• Add 5 µL of CellBlox Plus Blocking Buffer per sample prior to combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample containing 10³–10⁸ cells.

NovaFluor conjugates utilize Phiton™ technology, based on novel nucleic acid dye structures engineered to minimize spillover and spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein belonging to the selectin family of adhesion receptors. It consists of:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on all leukocytes including lymphocytes (except some memory T cells), monocytes, and polymorphonuclear cells.
