Product Specific Information

Description:
The DREG-56 monoclonal antibody reacts with human CD62L, a 76 kDa member of the selectin family. CD62L is expressed by neutrophils, monocytes, and subsets of T, B, and NK cells. It binds to glycosylated, fucosylated, and sulfated sialylated glycoproteins such as CD34, glycam-1, and MAdCAM-1, mediating lymphocyte rolling on activated endothelium and homing to high endothelial venules (HEV) of peripheral lymphoid tissues.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported:

• Flow cytometric analysis

Applications Tested:

• Pre-titrated and tested by flow cytometric analysis of normal human peripheral blood cells
• Recommended use: 5 µL (1 µg) per test
• A test = amount of antibody staining a cell sample in 100 µL final volume
• Cell number: 10⁵–10⁸ cells/test (empirically determined)

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for NovaFluor dyes.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample before combining with cells.
• For single-color controls: add 5 µL per 100 µL of cell sample (10³–10⁸ cells).

NovaFluor conjugates utilize Phiton™ technology, based on nucleic acid dye structures engineered for optimized fluorescent signatures and reduced spillover/spread.

Target Information

Human CD62L (L-Selectin) is a 74–95 kDa glycoprotein adhesion receptor. It contains:

• C-type lectin binding domain
• EGF-like domain
• Two short consensus repeat (SCR) sequences
• Spacer, transmembrane, and cytoplasmic regions

CD62L is constitutively expressed on most leukocytes, including lymphocytes, monocytes, and polymorphonuclear cells, except for many memory T cells.
