Target Information
Tight junctions are specialized regions of cell-cell contact that are particularly abundant in luminal epithelial cell sheets. In electron micrographs, tight junctions appear as belt-like bands of sealing strands that encircle each cell, functioning as a molecular gasket to prevent ion and molecule leakage. Beyond this barrier role, tight junctions maintain epithelial polarity by blocking diffusion of membrane proteins between apical and basolateral surfaces.
Proteins associated with tight junctions include ZO-1, ZO-2, ZO-3, cingulin, Rab-3b, and symplekin. Claudin-1 and Claudin-2 are key structural components of tight junction strands, inducing networks of strands characteristic of true tight junctions. Claudin-2 also functions as a paracellular channel with cation (Na⁺) selectivity and is primarily expressed in liver and kidney tissues.
