Target Information

ERN1 (serine/threonine kinase) or endoplasmic reticulum to nucleus signaling 1 is a human homologue of the yeast Ire1 gene product, possessing intrinsic kinase and endoribonuclease activity. It plays a key role in ER-based stress signaling, controlling IRE1 proteolysis in mammalian cells and being essential for fetal viability.
IRE1 acts as a sensor of unfolded proteins in the ER, initiating signal transduction to the nucleus by splicing XBP1 mRNA to produce a potent transcriptional activator of the unfolded protein response.
Null mutants are viable but essential under stress conditions that cause unfolded protein accumulation in the ER. Expression of a dominant negative form of IRE1p leads to apoptosis under such stress conditions.
