Target Information
EGFR (Epidermal Growth Factor Receptor, HER1, ErbB1)는 인간 염색체 7에 위치한 EGFR 유전자에 의해 암호화됩니다.
EGFR은 HER/ERbB 단백질 패밀리에 속하며, ERbB2, ERbB3, ERbB4와 함께 수용체 티로신 키나아제 역할을 합니다.
EGF 및 TGF-α 등의 리간드가 결합하면 수용체가 이합체화(dimerization)되고, 세포질 도메인 내 티로신 잔기의 자가 인산화가 유도됩니다.
EGFR 활성화는 Ras-ERK, PI3K-Akt, Jak-STAT, PKC 등 다양한 신호 경로를 통해 세포 성장 및 증식을 조절합니다.
Y1086 잔기 인산화는 GRB2 단백질의 결합을 유도하며, MAPK 경로 활성화를 촉진합니다.
활성화된 EGFR은 세포 내로 내포(endocytosis)되어 분해 또는 재활용됩니다.
