Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, a hormone critical for reproductive events such as the establishment and maintenance of pregnancy.
PR exists as two forms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), produced via alternative translation initiation sites.

• PR-B: Transcriptionally active form responsible for maintaining the endometrium and pregnancy, and inhibiting ovulation.
• PR-A: Identical to PR-B except for a 165 amino acid deletion at the N-terminus, which exposes a 140 amino acid inhibitory domain (ID) that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex including heat shock proteins and immunophilins. Upon progesterone binding, the receptor undergoes conformational changes, dimerizes, and binds to progesterone response elements (PRE), activating gene transcription.
A null mutation in the PGR gene results in pleiotropic reproductive abnormalities.
