Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy.
PR exists as a 94 kDa protein (Form A) and a 120 kDa protein (Form B) due to alternative translation initiation sites. PR‑B is transcriptionally active, responsible for maintaining the endometrium, pregnancy, and inhibiting ovulation.
PR‑A differs from PR‑B by a 165 amino acid deletion at the N‑terminus, exposing an inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Binding of progesterone induces conformational change, receptor dimerization, and binding to progesterone response elements (PRE), activating transcription.
A null mutation in the PGR gene leads to pleiotropic reproductive abnormalities.
