Target Information
The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events associated with pregnancy establishment and maintenance.
PR exists as two major isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), generated by alternative translation initiation sites. PR-B is transcriptionally active and responsible for activating genes related to endometrial maintenance, pregnancy, and ovulation inhibition. PR-A is identical to PR-B except for a 165 amino acid deletion at the N-terminus, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.
In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon binding to progesterone, the receptor undergoes a conformational change, dimerizes, and binds to progesterone response elements (PRE), activating transcription. Null mutations in the PGR gene can lead to pleiotropic reproductive abnormalities.
