Target Information

The progesterone receptor (PR) is a member of the steroid family of nuclear receptors. It mediates the physiological effects of progesterone, which plays a central role in reproductive events such as the establishment and maintenance of pregnancy.
PR exists as two isoforms: a 94 kDa protein (Form A) and a 120 kDa protein (Form B), resulting from alternative translation initiation sites.

  • PR-B: Transcriptionally active form, responsible for gene activation related to endometrial maintenance, pregnancy, and ovulation inhibition.
  • PR-A: Lacks 165 amino acids at the N-terminus compared to PR-B, exposing a 140 amino acid inhibitory domain that represses steroid hormone transcriptional activity.

In its inactive state, PgR forms a multiprotein complex with heat shock proteins and immunophilins. Upon binding progesterone, the receptor undergoes a conformational change, enabling dimerization and binding to progesterone response elements (PRE) to activate transcription.
A null mutation in the PGR gene can lead to multiple reproductive abnormalities.
