Target Information
Arginase-1 (Arg1)은 L-arginine을 urea와 L-ornithine으로 전환하는 35 kDa 효소로, 요소 회로의 마지막 단계를 담당합니다. 생성된 polyamine은 세포 증식 및 단백질 분해로 인한 독성 제거에 중요합니다. Arginase 1은 nitric oxide 합성효소의 기질인 arginine을 분해하여 NO 생성을 억제합니다.
인간과 마우스 모두에서 Arginase 1은 간, 호중구, myeloid-derived suppressor cells (MDSC), 신경 줄기세포 등에 발현됩니다. 인간에서는 혈액 호중구에는 발현되지만 CCR3+ 과립구에서는 발현되지 않는 것으로 보고되었습니다.
마우스에서 Arginase 1 발현은 대체적으로 활성화된 대식세포(M2a)의 특징 중 하나입니다. Arginase-1은 종양 침윤성 골수세포에서 발현될 수 있으며, 대부분의 간세포암에서 발견됩니다.
Arginase 1의 결함은 고암모니아혈증을 특징으로 하는 상염색체 열성 질환인 argininemia의 원인이 됩니다.
