Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. CD25 alone binds IL-2 with low affinity but forms a high-affinity receptor when associated with CD122 (IL-2Rβ) and CD132 (γ chain). The PC61.5 antibody inhibits IL-2 binding to both high and low affinity receptors, playing a role in lymphocyte differentiation, activation, and proliferation.

Applications Tested: Tested on mouse splenocytes or Con A-stimulated splenocytes at ≤0.5 µg/test (100 µL final volume). Optimal titration is recommended.

Target Information

CD25 (IL2RA) encodes the IL-2 receptor alpha chain, part of a heterotrimeric complex with IL2RB and IL2RG forming the high-affinity IL-2 receptor. Homodimeric IL2RA chains form low-affinity receptors, while IL2RB homodimers form medium-affinity receptors. IL2RA expression in mature thymocytes is monoallelic, regulating precise gene expression. IL2, mainly produced by mature T cells, functions as a growth, differentiation, and survival factor, stimulating B cell proliferation, NK cell activity, and inhibiting granulocyte macrophage colony formation. Mutations in IL2RA are associated with interleukin-2 receptor alpha deficiency and immune dysregulation.
