Product Specific Information

Description:
The CD28.2 monoclonal antibody reacts with the human CD28 molecule, a 44 kDa homodimer expressed by thymocytes, mature T cells, and plasma cells. CD28 is a ligand for CD80 (B7-1) and CD86 (B7-2), serving as a potent co-stimulator of T cells. Signaling through CD28 enhances IL-2 and IL-2 receptor expression and cytotoxicity of CD3-activated T cells.

Applications Reported:

• Flow cytometric analysis
• Immunoprecipitation (~2–5 µg per million cells)
• Immunohistochemical staining
• Functional assays for T-cell costimulation (use Functional Grade purified CD28.2, Product #16-0289-81)

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume). Cell number range: 10⁵–10⁸ cells/test. Optimize titer for best results.

Target Information

CD28 antigen is a 44 kDa disulfide-linked homodimeric T-cell surface glycoprotein, belonging to the immunoglobulin superfamily. It provides the critical second activation signal to T cells by binding CD80/CD86 on antigen-presenting cells. CD28 prevents T cells from entering anergic states or premature apoptosis. Expressed on most peripheral blood T lymphocytes and thymocytes, with variable levels across maturation stages.
Dysfunction of CD28 is associated with diseases such as mycosis fungoides and Sezary’s disease.
