Product Specific Information

Description:
The 1B1 monoclonal antibody reacts with mouse CD1d, a 48 kDa glycoprotein with structural homology to MHC class I molecules. CD1d associates with β2-microglobulin and functions similarly to MHC class II. 1B1 detects CD1d at varying levels on mouse leukocytes and recognizes β2m-associated CD1d.

Applications Reported:
Reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of mouse splenocytes. Use ≤0.8 µg per test (100 µL final volume). Optimal cell number ranges from 10⁵–10⁸ cells/test. Titrate antibody for best performance.

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assays.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL of CellBlox Plus per sample prior to combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information

CD1d is a heavy chain associated with β2-microglobulin on cortical thymocytes. It can also be expressed independently on human intestinal epithelial cells. CD1d belongs to the CD1 family of MHC-like glycoproteins that present lipid and glycolipid antigens to T cells. The CD1D gene encodes a divergent member of this family, forming heterodimers with β2-microglobulin. CD1 proteins differ in localization and lipid ligand specificity. CD1d localizes to late endosomes and lysosomes via a tyrosine-based cytoplasmic motif.
Diseases associated with CD1D include Mycobacterium malmoense infection and autoimmune diseases.
